At the 2019 Greek National Open Championship, Makrygiannis set four personal best times, and at the 2018 Greek Winter National Championships, he advanced to the final in four events. He has competed for Greece at the FINA World Championships and the LEN European Aquatic Championship.

Makrygiannis placed 42nd in the 100-yard back and 37th in the 200-yard back at the NCAA Championships (46.74).

His girlfriend Yusra could be seen cheering him on from the stands and offering advice to help him improve during other events, but he did not compete for USC during the 2020–21 season.
